UNDER THE MENTORSHIP OF BIOLOGY AND GEOLOGY FACULTY, OCCIDENTAL UNDERGRADUATE STUDENTS WILL GAIN VALUABLE EXPERIENCE WORKING WITH THIS MULTIBEAM SONAR SYSTEM, MANAGING AND MANIPULATING LARGE DATA SETS, AND ASSOCIATED MODELING, WHICH ARE KEY TO THEIR SUCCESS IN POST-GRADUATE CAREERS IN STEM FIELDS. IN ADDITION TO THE PHYSICAL EQUIPMENT, THIS AWARD WILL PROVIDE FUNDING FOR SOFTWARE LICENSES, IN-PERSON TRAINING, AND TIME FOR A RESEARCH ASSOCIATE WHO WILL DEPLOY, OPERATE, MAINTAIN, AND MANAGE THE SYSTEM AND ALL ASSOCIATED BATHYMETRIC DATA. THE LONG-TERM GOAL IS TO UNDERSTAND GAS SURVIVAL AND PERSISTENCE IN MACROPHAGES TO PROVIDE A CELL- TARGETED BASIS FOR IMPROVED TREATMENT FOR INFECTION. TOWARD THE LONG-TERM GOAL, THE OBJECTIVE IN THIS APPLICATION IS TO EXAMINE MACROPHAGE SECOND-LINE RESPONSES TO GAS FOLLOWING THE FAILURE OF LYSOSOMES TO ELIMINATE BACTERIA. THE CENTRAL HYPOTHESIS IS THAT GAS-INDUCED LYSOSOMAL PERMEABILIZATION ENABLES BACTERIAL AND LYSOSOMAL PROTEINS TO ENTER THE CYTOSOL, WHICH CONTINUES TO ALTER MACROPHAGE CELLULAR PROCESSES, INCLUDING LIMITING THE OXIDATIVE BURST RESPONSE AND INDUCING HISTONE RELEASE AND NON-LYTIC EXOCYTOSIS OF GAS. GUIDED BY PRELIMINARY DATA PRODUCED BY THE APPLICANT’S LABORATORY, THIS HYPOTHESIS WILL BE EXPLORED IN THREE SPECIFIC AIMS. THE WORK IN AIM 1 WILL DETERMINE WHETHER GAS NADASE IN THE CYTOSOL IMPAIRS THE OXIDATIVE BURST RESPONSE. MEASUREMENTS OF REACTIVE OXYGEN SPECIES, CELLULAR ENERGY POOLS, AND NADPH OXIDASE COMPLEX ASSEMBLY WILL DETERMINE WHETHER THIS BACTERIAL ENZYME NEGATIVELY IMPACTS THE MACROPHAGE OXIDATIVE BURST RESPONSE. AIM 2 WILL EXPLORE WHETHER GAS-INDUCED DAMAGE OF THE NUCLEAR ENVELOPE CAUSES DNA-FREE HISTONE RELEASE AND THE EXTENT TO WHICH THESE HISTONES HAVE ANTIMICROBIAL ACTIVITY AGAINST GAS IN THE MACROPHAGE CYTOSOL. PRELIMINARY DATA DEMONSTRATE THAT MACROPHAGES EXPEL GAS IN EXOSOMES AND THE WORK IN AIM 3 WILL CHARACTERIZE THESE GAS- CONTAINING EXOSOMES. TIME-LAPSE MICROSCOPIC OBSERVATIONS WILL BE COMPLEMENTED BY ELECTRON MICROSCOPY AND PROTEOMICS ANALYSES OF ISOLATED EXOSOMES. THE OUTCOME OF THIS WORK WILL INFORM APPROACHES AIMED AT RESTORING OR ENHANCING HOST CELL FUNCTION, WHICH BOTH AVOIDS THE PROBLEMS OF BROAD-SPECTRUM ANTIBIOTIC TREATMENT AND ENHANCES THE NATURAL ABILITY OF MACROPHAGES TO ERADICATE GAS. THE PROPOSED RESEARCH IS INNOVATIVE BECAUSE UNDERSTANDING THESE SECOND-LINE RESPONSES, WHICH ARE INTERMEDIATE PROCESSES BETWEEN LYSOSOMAL FAILURE AND DISEASE PATHOLOGY, WILL PROVIDE NEW DRUGGABLE TARGET PROTEINS AND PATHWAYS. THE CONTRIBUTION WILL BE A SIGNIFICANT FIRST STEP TOWARD DEVELOPING PHARMACOLOGICAL STRATEGIES THAT PREVENT, RATHER THAN TREAT SEVERE INVASIVE DISEASE AND REFRACTORY INFECTIONS.

ON THE OTHER HAND, THE REACTION OF LESS STABILIZED NUCLEOPHILES (CONJUGATE ACID PKA OF > 25) IS LESS WELL DOCUMENTED, IN PART DUE THE ENERGETIC BARRIERS ASSOCIATED WITH PROMOTING THEIR DECARBOXYLATION. IN CONTRAST, REPORTS THAT DETAIL THE FORMATION OF THE REGIOISOMERIC CYCLOPROPANATION PRODUCT REMAIN RELATIVELY SCARCE. TO ADDRESS THESE LIMITATIONS, WE HAVE DEVISED A NEW APPROACH THAT RELIES ON THE USE OF AROMATIC ANIONS AS A MEANS TO STABILIZE AND ENGAGE OTHERWISE RECALCITRANT NUCLEOPHILES IN DECARBOXYLATIVE COUPLING CHEMISTRY. SPECIFICALLY, WE RECENTLY DEMONSTRATED THAT ALLYL ESTER-SUBSTITUTED PHTHALIDES UNDERGO EFFICIENT ALLYLATION IN THE PRESENCE OF A PD/XANTPHOS CATALYST, DELIVERING A VARIETY OF FUNCTIONALIZED PHTHALIDES IN EXCELLENT YIELD. INTERESTINGLY, WHEN XANTPHOS IS REPLACED WITH THE BISAMINO LIGAND TETRAMETHYLETHYLENEDIAMINE (TMEDA), A CYCLOPROPANATED PHTHALIDE IS ISOLATED IN HIGH REGIOSELECTIVITY. THE GOAL OF AIM 1 IS TO SIGNIFICANTLY EXPAND ON THESE PRELIMINARY FINDINGS BY APPLYING OUR OPTIMAL PD/XANTPHOS DAA CATALYST TO ACCESS A VARIETY OF BIOLOGICALLY RELEVANT MOTIFS, INCLUDING FUNCTIONALIZED ISOINDOLINONE AND FLUORENE DERIVATIVES. ALTERNATIVELY, AIM 2 SEEKS TO EXPLOIT THE REACTIVITY PD/TMEDA-BASED CATALYST TO DEVELOP A GENERAL DECARBOXYLATIVE CYCLOPROPANATION OF PHTHALIDE-BASED NUCLEOPHILES. PREVIOUS WORK HAS PROPOSED THAT PAST EPISODIC FRESHWATER FLUXES KNOWN AS HEINRICH EVENTS IN THE NORTHERN HEMISPHERE ABRUPTLY MODULATE TROPICAL HYDROCLIMATE IN REGIONS LIKE THE INDIAN PACIFIC WARM POOL. IN THIS PROJECT, CAVE DEPOSITS FROM THE PHILIPPINES ARE USED TO RECONSTRUCT PAST HYDROLOGIC CHANGE AND INVESTIGATE THE HYDROLOGIC RESPONSE OVER THE INDIAN PACIFIC WARM POOL TO HEINRICH EVENTS. THE PHILIPPINES IS IDEALLY LOCATED FOR THIS WORK AS IT IS SITUATED WITHIN THE INDIAN PACIFIC WARM POOL AND IS SENSITIVE TO SEASONAL VARIATIONS IN RAINFALL ASSOCIATED WITH THE ASIAN MONSOON SYSTEMS. THIS WORK WILL ACCOMPLISH THREE PRIMARY TASKS: FIRST, IT WILL EXPAND CURRENT CAVE MONITORING EFFORTS IN THE PHILIPPINES TO ENCOMPASS A MULTI-YEAR, MULTI-CAVE MONITORING NETWORK TO BETTER CONSTRAIN SEASONAL VARIATIONS IN KARST HYDROLOGY. SECOND, IT WILL CONTRIBUTE NEW HIGH TEMPORAL RESOLUTION CAVE GEOCHEMISTRY RECORDS SPANNING NUMEROUS HEINRICH EVENTS. THIRD, GEOCHEMICAL DATA-MODEL COMPARISONS WILL BE CONDUCTED TO EVALUATE THE MECHANISMS DRIVING GEOCHEMICAL VARIABILITY IN THESE RECORDS DURING HEINRICH EVENTS. BY BUILDING CAPACITY FOR DATA COLLECTION AND CLINICAL INTERVENTION, INCREASING OPPORTUNITIES FOR PEER EDUCATION AND STUDENT-LED PREVENTION EFFORTS, AND EXPANDING TRAINING FOR SUICIDE-PREVENTION AND REFERRAL SKILLS-BUILDING ACROSS CAMPUS, WE WILL ADDRESS STUDENT NEEDS AND BUILD ROBUST PREVENTION, IDENTIFICATION, AND REFERRAL SUPPORT RELATED TO MENTAL AND SUBSTANCE USE DISORDERS AND SUICIDALITY AMONG OXY STUDENTS. THE PROJECT WILL TAKE AN EVIDENCE-BASED, PUBLIC HEALTH APPROACH TO ADDRESS THE NEEDS OF ALL STUDENTS, WITH A FOCUS ON FIRST-YEAR (27%), STUDENTS OF COLOR (46%), AND LGBTQIA+ IDENTIFYING (46%) STUDENT POPULATIONS, AS WELL AS STUDENTS IN OXY'S STUDY ABROAD AND INTERNATIONAL PROGRAMS, ALL OF WHOM ARE AT GREATER RISK OF NEGATIVE OUTCOMES FROM SUBSTANCE MISUSE, MENTAL HEALTH DISTRESS, AND HAVE REPORTED FEELING LESS SENSE OF CONNECTEDNESS AND BELONGING THAN THEIR COUNTERPARTS ON CAMPUS. THE OVERARCHING GOALS OF THIS PROJECT ARE TO INCREASE HELP-SEEKING BEHAVIORS FOR MENTAL HEALTH AND SUBSTANCE USE ISSUES, TO ENHANCE BELONGING AND CONNECTEDNESS AMONG STUDENTS, AND TO REDUCE SUICIDE AND MENTAL AND SUBSTANCE USE DISORDERS. THERE ARE FOUR MAIN OBJECTIVES: 1. USING A TRAUMA-INFORMED LENS, ENHANCE CLINICAL CAPACITY TO IDENTIFY AND INTERVENE WITH STUDENTS AT RISK FOR SUICIDE, DEPRESSION, SERIOUS MENTAL ILLNESS/SERIOUS EMOTIONAL DISTURBANCES, AND/OR SUBSTANCE MISUSE, AND CONNECT STUDENTS TO TREATMENT AND RECOVERY RESOURCES WHEN NEEDED. -EMMONS WELLNESS CENTER (EMMONS) WILL IMPLEMENT AN SBIRT (SCREENING, BRIEF INTERVENTION, AND REFERRAL TO TREATMENT) MODEL, TRAIN CLINICAL STAFF IN BRIEF MOTIVATIONAL INTERVIEWING, AND INTEGRATE DAST-10 AND CCAPS-36 SCREENING TOOLS DURING CLINICAL VISITS. SOME OF THIS SEDIMENT IS DEPOSITED IN DEEP OCEANIC TRENCHES AND SUBSEQUENTLY SUBDUCTED BENEATH THE CONTINENTS. BECAUSE THESE SEDIMENTS ARE NOT DENSE ENOUGHT TO SINK INTO THE MANTLE, MUCH OF THE SUBDUCTED SEDIMENT IS THOUGHT TO BECOME RE-INCORPORATED INTO THE CONTINENTS. THIS RE-ADDITION OF SUBDUCTED SEDIMENT TO THE BASE OF CONTINENTS CAN HELP UNDERSTANDING OF THE PATTERNS OF DEFORMATION AND THE DISTRIBUTION OF ELEMENTS WITHIN THE CONTINENT. IT CAN POTENTIALLY REDUCE THE STRENGTH OF TECTONIC PLATES, SO THAT SOME ZONES MAY BE MORE PRONE TO FAULTING AND SEISMIC ACTIVITY. INTRODUCING SEDIMENT INTO THE LITHOSPHERE CAN ALSO RECHARGE THE ELEMENTS THAT HAVE PREVIOUSLY BEEN DEPLETED BY TECTONIC PROCESSES, INCLUDING CRITICAL MINERALS. ONE OF THE FUNDAMENTAL CHALLENGES THIS RESEARCH ADDRESSES IS DETERMINING THE DISTRIBUTION AND GEOMETRY OF SUBDUCTED SEDIMENT IN THE CONTINENTAL CRUST. THIS PROJECT ESTABLISHES A NEW COLLABORATION AND PROVIDES SUPPORT FOR TWO EARLY-CAREER RESEARCHERS AND FIVE UNDERGRADUATE STUDENTS FROM HISTORICALLY MARGINALIZED AND UNDERREPRESENTED GROUPS AT TWO DIFFERENT UNIVERSITIES. THE STUDENTS RECEIVE TRAINING IN A VARIETY OF GEOLOGIC FIELD METHODS, CONDUCT CHEMICAL AND ISOTOPIC ANALYSES AT MULTIPLE UNIVERSITY LABORATORIES OUTSIDE OF THEIR HOME INSTITUTIONS, AND DEVELOP EFFECTIVE SCIENCE COMMUNICATION SKILLS. END-MEMBER MODELS FOR THE GEOMETRY OF SUBDUCTED SEDIMENT AT DEPTH INCLUDE REPLACEMENT OF THE CRUSTAL MATERIALS WITH UNIFORM, SHEET-LIKE LAYERS OF SEDIMENT AND NON-UNIFORM, DIAPIR-LIKE BODIES DISTRIBUTED THROUGHOUT THE CRUST. THIS RESEARCH WILL TEST THESE MODELS IN SOUTHERN CALIFORNIA AND ARIZONA, WHICH EXPERIENCED WIDESPREAD SEDIMENT SUBDUCTION AND ACCRETION DURING THE LATE CRETACEOUS TO EARLY PALEOGENE. THE SUBDUCTED SEDIMENT, LOCALLY CALLED THE PELONA-OROCOPIA-RAND SCHIST (?OROCOPIA SCHIST?), HAS BEEN TECTONICALLY EXHUMED TO THE SURFACE ALLOWING FOR DIRECT INVESTIGATION INTO ITS STRUCTURAL AND CHEMICAL PROPERTIES. THE OROCOPIA SCHIST HAS A UNIQUE COMBINATION OF JUVENILE RADIOGENIC ISOTOPIC COMPOSITIONS AND HEAVY STABLE OXYGEN ISOTOPE RATIOS, WHICH DISTINGUISHES IT FROM OTHER MAJOR ISOTOPIC RESERVOIRS IN THE AREA. RECENT STUDIES AND PRELIMINARY DATA INDICATE THAT IGNEOUS ROCKS THAT TRANSVERSED THE CRUST DURING THE MID-CENOZOIC IGNIMBRITE FLARE-UP IN AREAS WHERE THE OROCOPIA SCHIST IS PRESENT RECORD THE UNIQUE ISOTOPIC SIGNATURE, WHEREAS MAGMATISM OUTSIDE OF THIS AREA DOES NOT. ADDITIONALLY, IGNEOUS ROCKS THAT INTRUDED INTO THE CRUST PRIOR TO ACCRETION OF THE OROCOPIA SCHIST DO NOT FEATURE THE UNIQUE ISOTOPIC FINGERPRINT. DESPITE THEIR SMALL SIZE, CIRQUE GLACIERS PLAY AN OUTSIZED GEOMORPHIC AND ECOLOGICAL ROLE IN ALPINE LANDSCAPES, LARGELY THROUGH REGIONALLY VARIABLE CHANGES IN HYDROLOGY AND SEDIMENT TRANSPORT. UNDERSTANDING HOW THESE GLACIERS RESPONDED TO PAST CLIMATE CHANGES AND HOW THEY WILL RESPOND TO FUTURE CLIMATE CHANGES IS PREREQUISITE FOR UNDERSTANDING THE PAST EVOLUTION AND FUTURE FATE OF ALPINE CATCHMENTS IN THE WESTERN U.S. AND ELSEWHERE. THIS PROJECT WILL ADDRESS THIS NEED BY DEVELOPING DETAILED GLACIOLOGICAL DATASETS, BOTH MODERN AND PALEO, FROM AN EXCEPTIONALLY WELL-CONSTRAINED STUDY SITE IN THE TETON RANGE, WYOMING, AND INTEGRATING THEM WITH A STATE-OF-THE-ART MODEL TO ACCURATELY REPRESENT GLACIERS AS THEY SHRINK AND DISAPPEAR. ULTIMATELY, THIS WORK WILL PRODUCE INSIGHT INTO THE PAST, PRESENT, AND FUTURE ROLE OF GLACIERS AS AGENTS OF ALPINE LANDSCAPE EVOLUTION WHILE DEVELOPING AN OPEN-SOURCE GLACIER MODEL, WHICH WILL BE APPLICABLE TO GLACIAL SETTINGS GLOBALLY. WITH A CONTINUOUS RECORD OF GLACIER EXTENT, THICKNESS, AND VOLUME THIS WORK WILL LAY THE FOUNDATION FOR FUTURE STUDIES, BOTH GEOMORPHIC AND ECOLOGICAL IN SCOPE, THAT QUANTIFY THE IMPACT OF GLACIER CHANGE ON ALPINE LANDSCAPES FACING COMPLETE DEGLACIATION. THIS PROJECT WILL COMPILE DATA FROM CAVE DEPOSITS, CORALS AND TREE RINGS, AS WELL AS DEVELOP NEW RECORDS FROM CAVES IN THE PHILIPPINES AND NORTHERN AUSTRALIA, TO PRODUCE A RECONSTRUCTION OF THE BEHAVIOR OF THE AUSTRAL-ASIAN MONSOON FOR THE LAST 1000 YEARS. THIS RECONSTRUCTION WILL GUIDE A SET OF CLIMATE MODEL SIMULATIONS TO IDENTIFY THE DRIVERS OF MONSOON VARIABILITY. THE RESULTS WILL IMPROVE DECADAL PREDICTION THE AUSTRAL-ASIAN MONSOON. THE PROJECT WILL SUPPORT THE PARTICIPATION OF A POSTDOC AND UNDERGRADUATE STUDENTS IN THE RESEARCH, AN ART-SCIENCE COLLABORATION, K-12 EDUCATION, AND PUBLIC OUTREACH TO PRIMARY AND SECONDARY STUDENTS IN IOWA, CALIFORNIA, NEW MEXICO AND NORTHERN AUSTRALIA. ONE OF THE LONGEST-HELD MODELS INVOKES A SHALLOW SUBDUCTING OCEANIC PLATE BENEATH THE MOJAVE DESERT TO WYOMING TO LARGE-SCALE MOUNTAIN BUILDING AS FAR EAST AS THE ROCKY MOUNTAINS. THIS STUDY TESTS THE VIABILITY OF THE FLAT SLAB MODEL AGAINST ALTERNATIVE HYPOTHESES, INCLUDING THE COLLISION AND TRANSLATION OF A FAR-TRAVELED TERRANE, THROUGH DETAILED STUDY OF THE PELONA-OROCOPIA-RAND SCHIST. THIS SUITE OF ANCIENT METAMORPHIC ROCKS SPAN FROM CALIFORNIA TO ARIZONA, A CRITICAL LOCATION TO DISTINGUISH BETWEEN THE TECTONIC MODELS. THE RESEARCHERS WILL TEST PREDICTIONS ABOUT THE LOCATION, CONDITIONS, AND TIMING OF TECTONIC EVENTS DERIVED FROM STANDARD RECYCLING OF OCEANIC CRUST, FLAT SLAB SUBDUCTION, AND TERRANE COLLISION BY DETERMINING THE AGE, PRESSURE, AND TEMPERATURE RECORDED BY THE METAMORPHIC MINERAL GARNET IN THE PELONA-OROCOPIA-RAND SCHIST. THE PROJECT WILL PROMOTE THE DEVELOPMENT OF TWO EARLY-CAREER WOMEN IN STEM, FOSTER NEW COLLABORATIONS ACROSS R1, R2, AND PUI CAMPUSES, FACILITATE THE TRAINING OF GRADUATE AND UNDERGRADUATE STUDENTS FROM UNDERREPRESENTED GROUPS, AND SUPPORT THE DEVELOPMENT OF ANALYTICAL FACILITIES.
