THROUGH THE FELLOWSHIP, THE PI AIMS TO STUDY THE EFFECTS OF CHANGING CLIMATE ON GREENHOUSE GAS (GHG) EMISSIONS IN WETLAND ECOSYSTEMS. THE MAIN GOAL OF THIS FELLOWSHIP IS TO BETTER UNDERSTAND THE DYNAMICS OF GHGS WITHIN WETLAND ECOSYSTEMS BY MEASURING GHG FLUXES IN MINERAL SOIL FORESTED WETLANDS THAT ARE NOT WELL STUDIED. THE PROJECT WILL AIM TO IMPROVE THE DETECTION OF GHG EMISSIONS FROM WETLANDS USING SATELLITES, REDUCE UNCERTAINTIES IN THE GHG FLUXES ACROSS LARGER SPATIAL EXTENTS, AND VALIDATE SATELLITE-BASED GHG EMISSION DATA. THE PROJECT DESIGN ALLOWS FOR THE QUANTIFICATION OF GHG FLUXES FROM ALL MAJOR POOLS (I.E., SOILS, STEMS, KNEES), AND FOR THE INVESTIGATION OF SOIL-VEGETATION INTERACTIONS. THE FELLOWSHIP WILL STRENGTHEN THE PRINCIPAL INVESTIGATOR?S ABILITY TO ATTRACT DIVERSE STUDENTS (WOMEN, FIRST-GENERATION, AND MINORITY STUDENTS) TO THE FIELD AND PROVIDE THEM WITH OPPORTUNITIES TO DEVELOP THE RESEARCH SKILLS THAT CAN LEAD THEM TO SUCCESSFUL FUTURE CAREERS. IN THIS PROJECT, THE PI WILL EXTEND KNOWLEDGE OF CARBON DYNAMICS BY ESTABLISHING A NEW SITE FOR MEASURING GHGS FLUXES IN MINERAL SOIL FORESTED WETLANDS THAT ARE NOT WELL STUDIED. THE OVERARCHING OBJECTIVES ARE TO (1) IMPROVE UNDERSTANDING OF THE CONTROLS ON GHG FLUXES IN MINERAL SOIL WETLANDS, AND (2) TO BETTER UNDERSTAND THE EFFECTS OF LANDSCAPE POSITION AND FOREST COMPOSITION ON THE GHG FLUXES BETWEEN TERRESTRIAL ECOSYSTEMS AND THE ATMOSPHERE. CURRENTLY, LITTLE IS KNOWN ABOUT THE SPECIFIC ROLES GLIS3 PLAYS DURING DEVELOPMENT OR HOW DIVERGENCE FROM ITS NORMAL FUNCTION PROMOTES DISEASE AT THE MOLECULAR LEVEL. IN THIS PROPOSAL, EXPERIMENTS ARE DESCRIBED INTENDED TO CHARACTERIZE PANCREATIC AND RENAL DEVELOPMENT IN THE GLIS3EX4DEL/EX4DEL MUTANT ZEBRAFISH LINE THAT LACKS FUNCTIONAL GLIS3 EXPRESSION. IN SPECIFIC AIM 1, WE WILL CHARACTERIZE PANCREATIC DEVELOPMENT IN GLIS3 KNOCKOUT FISH TO DETERMINE WHAT ROLE GLIS3 PLAYS IN DIRECTING DEVELOPMENT OF THE PANCREATIC DUCTS AND THE SPECIFICATION OF SECONDARY ISLETS THAT EMERGE FROM THE DUCTAL EPITHELIUM. OUR CENTRAL HYPOTHESIS IS THAT DEVELOPMENTAL EXPOSURE TO LOW LEVELS OF PSYCHOACTIVE DRUGS THROUGH CONTAMINATED WATER SOURCES AFFECTS GENE EXPRESSION IN THE DEVELOPING NERVOUS SYSTEM WITH BEHAVIORAL CONSEQUENCES. THIS PROPOSED BASIC BIOMEDICAL RESEARCH WILL EXPOSE DEVELOPING ZEBRAFISH TO AN ENVIRONMENTALLY-RELEVANT COMBINATION OF PSYCHOTROPIC COMPOUNDS TO EVALUATE THE EFFECTS OF THIS FORM OF EXPOSURE ON GENE EXPRESSION AND FUNCTION OF NERVOUS SYSTEM. ABEFORE-AFTER CONTROL-IMPACT (BACI) STUDY DESIGN WILL BE USED TO ASSESS BIRD COMMUNITYCHANGES ACROSS 64 SYSTEMATICALLY PLACED SAMPLING LOCATIONS REPRESENTING BOTH MANAGED (N 19)AND CONTROL UNITS WITHIN CRNWR (N 45). ALTHOUGH THE CURRENT DATASET IS SKEWED TOWARDCONTROL UNITS, IT WILL ALSO SERVE AS BASELINE DATA FOR UNITS SCHEDULED FOR FUTURE MANAGEMENT,ENABLING FURTHER COMPARISONS OVER TIME. AUDIOMOTH BIOACOUSTIC RECORDERS WERE DEPLOYED IN2021 AND 2022 TO ESTABLISH PRE-TREATMENT BIRD COMMUNITY BASELINES. IN SPRING 2026, SURVEYS WILLBE CONDUCTED USING THE SAME PROTOCOL.
