THE ESTABLISHMENT OF A COMPACT X-RAY FREE ELECTRON LASER (CXFEL) FACILITY AT ARIZONA STATE UNIVERSITY WILL ENABLE THESE ADVANCED STUDIES AND FULFILL AN URGENT NEED IN THE BROAD U.S. RESEARCH COMMUNITY TO DIRECTLY OBSERVE, MANIPULATE, AND CONTROL QUANTUM DYNAMICS IN COMPLEX MATERIALS, MOLECULES, AND INTERFACES. THESE CAPABILITIES WILL REVEAL NOVEL BIOLOGICAL PROCESSES, ENABLE NEW ENERGY-EFFICIENT TECHNOLOGIES, AND DECODE QUANTUM BEHAVIOR THAT CAN DRIVE NEW COMPUTING METHODS AND NOVEL MATERIALS. A DIVERSE POPULATION OF UNDERGRADUATE AND GRADUATE STUDENTS WILL BE ENGAGED IN THE SCIENTIFIC AND PROJECT MANAGEMENT ASPECTS OF THIS MID-SCALE RESEARCH INFRASTRUCTURE DEVELOPMENT EFFORT. THE CXFEL WILL UNIQUELY OFFER FULL CONTROL OF THE X-RAY PHASE USING ELECTRON-BEAM PATTERNING THAT ALLOWS CUSTOMIZED TIME-STRUCTURE OF THE BEAM, E.G., ATTOSECOND PULSES, VERY NARROW LINEWIDTHS, AND EXTREMELY PRECISE TIMING AMONG MULTIPLE PULSES WITH DIFFERENT COLORS. MULTIPLE END-STATIONS WILL BE CONFIGURED TO SUPPORT X-RAY SPECTROSCOPY, SMALL- AND WIDE-ANGLE X-RAY SCATTERING, COINCIDENT ATTOSECOND MOMENTUM IMAGING, AND TIME-RESOLVED X-RAY CRYSTALLOGRAPHY FOR STUDIES IN BIOLOGY, QUANTUM MATERIALS, ATOMIC AND MOLECULAR SCIENCE, AND OPTICAL SCIENCES. THESE FINDINGS ARE LIKELY DUE TO INDIVIDUAL DIFFERENCES IN AEROBIC-FITNESS RESPONSES, LONG ESTABLISHED IN ADULTS USING PEAK OXYGEN CONSUMPTION (VO2PEAK) AND FIRST REPORTED IN OLDER ADULTS WITH ADRD BY OUR TEAM. AD/ADRD EXERCISE TRIALS REPORT LARGE VARIANCE IN VO2PEAK CHANGES FROM MODERATE-INTENSITY CONTINUOUS TRAINING (MICT). MECHANISTICALLY, ANIMAL STUDIES SUPPORT AEROBIC EXERCISE MODIFYING AD’S AT(N) BIOMARKERS (AMYLOID-BETA [ASS], TAU, AND NEURODEGENERATION), BUT SIMILAR HUMAN STUDIES ARE RARE. HENCE, PRECISION EXERCISE IS CRITICAL TO IDENTIFY MICT NON-RESPONSE EARLY TO INITIATE ALTERNATIVE INTERVENTIONS (HIGH INTENSITY INTERVAL TRAINING [HIIT] OR COMBINED AEROBIC AND RESISTANCE EXERCISE [CARE]). OUR CENTRAL HYPOTHESIS IS THAT MICT AUGMENTED WITH HIIT OR CARE WILL IMPROVE AEROBIC FITNESS, WHITE MATTER HYPERINTENSITY (WMH), AND PLASMA BIOMARKERS, WHICH UNDERLIE EXERCISE’S COGNITIVE EFFECTS. THIS TRIAL BUILDS ON OUR PREVIOUS WORK SHOWING: SUCCESSFUL RECRUITMENT, RETENTION, ADHERENCE, AND SAFETY; 6-MONTH MICT MAINTAINED MEMORY AND REDUCED WMH; INDIVIDUAL DIFFERENCES IN VO2PEAK AND COGNITIVE RESPONSES TO MICT; MICT IMPROVED PHYSICAL FUNCTION, QOL, AND CAREGIVER DISTRESS; PLASMA NEUROFILAMENT LIGHT CHAIN (NFL) PREDICTED COGNITION; AND MICT AFFECTED PLASMA P-TAU181. IT WILL RANDOMIZE 108 PARTICIPANTS 2:1 TO 3-MONTH MICT OR 6-MONTH STRETCHING CONTROL AFTER BASELINE. VO2PEAK WILL BE ASSESSED AFTER 3-MONTH MICT TO IDENTIFY NON- RESPONDERS (<5% INCREASE) AND RE-RANDOMIZE THEM 1:1 TO HIIT OR CARE FOR 3 MONTHS. RESPONDERS WILL CONTINUE MICT FOR 3 MONTHS. THIS PROJECT AIMS TO TAKE ADVANTAGE OF THE UNIQUE CHARACTERISTICS OF DNA SELF-ASSEMBLY, ALONG WITH ITS CHEMICAL AND STRUCTURAL PROPERTIES TO CREATE SYNTHETIC MATERIALS WITH PRECISELY-CONTROLLABLE ELECTRONIC AND MAGNETIC PROPERTIES WITH THE ULTIMATE GOAL OF DEVELOPING MAGNETIC TOPOLOGICAL INSULATORS (TIS). THE INTRODUCTION OF ROBUST MAGNETISM INTO TIS WILL ALLOW UNIQUE QUANTUM PHENOMENA TO BE REALIZED IN THE SOLID STATE THAT COULD BE EXPLOITED FOR APPLICATIONS INCLUDING QUANTUM COMPUTING, ELECTRONICS, SPINTRONICS, OPTOELECTRONICS, AND RENEWABLE ENERGY. ACCOMPLISHING THIS VISION WILL REQUIRE A CONVERGENCE BETWEEN CHEMISTRY, BIOLOGY, MATERIALS SCIENCE, ENGINEERING, NANOTECHNOLOGY, COMPUTATIONAL MODELING, AND CONDENSED MATTER PHYSICS. TO ACHIEVE CONVERGENCE, THIS PROJECT BRINGS TOGETHER A DIVERSE TEAM OF EXPERTS SPANNING THESE DISCIPLINES THAT WILL WORK COLLABORATIVELY TO CO-DESIGN AND CO-DEVELOP THESE SYSTEMS. DEVELOPING THE ABILITY TO PRECISELY CONTROL THE ELECTRONIC BAND STRUCTURE AND MAGNETIC PROPERTIES OF MATERIALS WILL HAVE BROAD IMPACTS ACROSS SOCIETY IN THE COMING DECADES. IN ADDITION, THIS PROJECT INCLUDES A FRAMEWORK FOR NUCLEATING A NEW SCIENTIFIC COMMUNITY FOCUSED ON UTILIZING THE UNIQUE SPATIAL AND TOPOLOGICAL PROPERTIES OF SOFT MATTER TO CREATE TOPOLOGICAL INSULATORS WITH PRECISELY TAILORED ELECTRONIC AND MAGNETIC PROPERTIES. THIS COMMUNITY WILL REPRESENT A CONVERGENCE OF THOUGHTS, LANGUAGE, AND IDEALS FROM A BROAD SPECTRUM OF THE SCIENCE AND ENGINEERING COMMUNITY. THIS PROJECT AIMS TO DEVELOP A FRAMEWORK THAT ENABLES PRECISION TOPOLOGICAL CONTROL OVER THE ELECTRONIC AND MAGNETIC PROPERTIES OF MATERIALS. ACHIEVING THIS VISION REQUIRES A CONVERGENCE BETWEEN COMPLEMENTARY DISCIPLINES TO ULTIMATELY FUSE THE REALMS OF SOFT MATTER, REAL-SPACE TOPOLOGY WITH MOMENTUM-SPACE (K-SPACE) TOPOLOGY. SPECIFICALLY, THE AIM IS TO UTILIZE THE UNIQUE PROGRAMMABILITY AND UNPARALLELED SPATIAL, STRUCTURAL, AND SELF-ASSEMBLY CAPABILITIES AFFORDED BY DNA NANOTECHNOLOGY TO PRECISELY TAILOR THE TOPOLOGY OF METAL IONS TO ULTIMATELY CREATE MAGNETIC TOPOLOGICAL INSULATORS THROUGH ITERATIVE, RATIONAL DESIGN. THE INTRODUCTION OF MAGNETISM INTO TOPOLOGICAL INSULATORS IS CURRENTLY A MAJOR UNRESOLVED CHALLENGE IN THE CONDENSED MATTER PHYSICS COMMUNITY; AND REAL-SPACE, DNA-BASED TOPOLOGICAL CONTROL PROVIDES A UNIQUE PATH TOWARD SOLVING THIS CHALLENGE. THE DEVELOPMENT OF ATOMICALLY PRECISE, METALATED DNA MOTIFS WILL ALLOW CONTROL OVER ELECTRONIC PROPERTIES, AND THE DESIGN OF PURPOSE-SPECIFIC, SELF-ASSEMBLED BAND STRUCTURE.
